Comparison of Various Storage Media for the Preservation of Neisseria gonorrhoeae

Abstract
To seek a inexpensive and simple method to preserve Neisseria gonorrhoeae, a few methods were compared. The authors also gave the method a trial the survival of the strains according to temperature during transit from transport media. The following results were obtained.
The following methods kept the culture alive for a week : tryptic soy agar with 15% glycerol at -20 *C : 10% skim milk at -201C , cystine tryptic agar at 351C .
Gonococci survived for more than 8 weeks in the one half strength dextrose starch agar and one half strength dextrose starch with VCN, but GC medium1/2 could be preserved for average 4.4 weeks.
It is recommended that preservation of gonococci in one half strength dextrose starch agar with VCN, which produces less frequent contamination is a practical to maintain cultures for teaching and quality control.
The survival period of gonococci with Stuart¢¥s transport media at 351C, room temperature and 4 C were retained for 24 hours respectively. Stuart¢¥s transport with 1 % charcoal survived for more than 4 days at room temperature and 4 C, but alive for 24 hours at 35¡É.
